•Proof of concept study to determine if simultaneous HPV E6,E7 mRNA and PD-L1 is possible on cervical cytology samples.

•Here we show dual E6,E7 and PD-L1 expression on the same sample. This information is currently not available in a single test by any other method.

•Based on this small sample set, PD-L1 expression decreases in high grade lesions. Additional samples should be tested to confirm this finding.
Thanks.

Don't know all of it but E6 and E7 are proteins that when overly expressed can significantly increase the risk of cervical cancer. PD-L1 are another group of proteins that work in the immune system to determine what to target and what to leave alone. Sometimes when people have cancer the PD-L1  in the cancerous cells tells the immune system not to attack the cancerous cells.

Overly expressed?  Does that mean they occur in cells at a higher rate than normal?

HPV is human papilloma virus, a sexually transmitted organism that can cause cancer in men and women. mRNA is mitchodrial ribonucleic acid. That's all I got.

Yeah, basically its making too many copies of the protein. I think the e-6 and e-7 are tied to regulation of the cell's life so it plays a role in the cancer cells becoming immortal.

HPV E6/E7 are genes within HPV that inactivate tumor suppressor proteins. Thus promoting tumor growth.

PD-L1 is a ligand protein that also seems to allow tumor growth when present

Perhaps your OP suggests using HPV E6/E7 to inactivate PD-L1... so that the body could then mount the proper immune response in its absence. And thus, a reduction in tumor size/growth.

I think this is partially correct. E6 and E7 are HPV proteins which promote cervical intrepithelial neoplasia converting to invasive cancer.  PD-L1 is a target for cancer immunotherapy, as it is a “checkpoint” between some cancer cells and our immune system.

A single assay which measures the risk for cancer progression (E6/E7) coupled with a marker for cancer therapy (PD-L1 inhibitors) would be of value for women with CIN, especially if it could be obtained from something like a Pap smear.

ETA It doesn’t appear PD-L1 inhibitors are FDA approved for cervical cancer but are for urothelial carcinoma, of which some subtypes may be promoted by HPV infection.
