The only ways to replenish talent over time in a dynasty league are:

a) draft good rookies

b) fleece people with trades

or

c) be lucky on grabbing the best waiver pickups

The second options is unreliable, and the third option only works well if your team is down in the standings (or you use blind bidding for free agents AND are sharper than your rivals).  So in short, the rookie draft is essential.

That said, you don't have to gain your value from it by actually drafting rookies.  You can also build and sustain a dynasty by making effective trades of high perceived-value picks for established players.  Best way to use the rook draft is the same as anything else...try to maximize your return on investment, whether by holding or dealing each asset.

How much value you put on them depends on whether you're in rebuilding or win-now mode.

Great post all the way around. You can't keep fleecing people on trades, they will stop trading with you.

I think the bolded is the best piece of advice.  Rookies are a crapshoot no matter what.  For every Zeke that performs as most think there are T-Richardson's or Laquon Treadwell's that bust.  I would much rather use the shiny new toy hype to trade rookie picks for established NFL players or players that have shown they can be productive if given the chance.  Getting players that are 2nd or 3rd guys that have shown glimpses and are now in new situations or new teams  that will get them more opportunity is the best way to stay on top in dynasty. 

Also sprinkling in productive older players that nobody wants that you can get cheap help round out your roster.  These players are always available as most dynasty owners put such a huge premium on the shiny new toys.  Acquiring proven vets for cheap has been a nice way to fly under the radar on the "fleecing people with trades" aspect.
