Best ball is when you do not name starters and every player on your roster is eligible to score. Your team score results from the highest scorers among your roster in accordance with the league's rules. Some leagues start for example. one QB, two RB, three WR, one TE, one PK and one DST. Others include a flex scorer or two among a variety of positions, typically RB/WR/TE.
