It's a pretty big deal considering it's never happened before. If it were Brady vs Brady I'm sure many would feel different.
Edit to add...I don't mean to diss the people who don't care about this game from a personal standpoint. We all have our opinions and likes/dislikes.
BUT IF YOU DON'T BELIEVE THAT THIS IS GREAT FOR THE NFL...then you aren't thinking this through.

Why would people be jealous of the Mannings but not Brady? No, I think a lot of people just don't like them. They're doofs, they choke away big games, and when it comes to Peyton, displaces blame when it clearly falls on him. To some people they are simply very unlikable. And when a player is praised unconditionally by the media at every oppurtunity, it can often make it worse.
Is this game good for the NFL? Sure. It has a "storyline". Of course it's going to get attention. But I don't think that means everybody who tunes in will do so because they love the Mannings -- I think many people will tune in to ridicule them and "dislike" them. Although, they'll both probably play very well, because after all, it is the regular season.
Like I said, our feelings on the Mannings are meaningless. But this world would be a better place if more families raised their kids like Archie and his wife did Peyton and Eli.As far as the reasons this game is VERY important to the NFL:
1) New TV Deal - the NFL broke the bank with it's new deal, with ESPN and NBC investing the bulk. A good start to SNF is important. I'm guessing the ratings will be higher than they have in a long time. I'm guessing companies looking to advertise feel the same way. I'm guessing NBC is reaping the benefits of that. That leads to $$ for everyone, and is the first step towards a successful deal for all and a BIGGER one down the road.
2) New Commish - Roger Goodell needs to get off on the right foot. This game's success will be the perfect start.
3) The new CBA - part of the new deal is that they are sharing more of the pie, which includes merchandise sales. That part of the pie needs to grow, and jerseys with Manning on the back will help that. The fact that the Giants and Colts are two teams that are in the lower half of the top 10 in total sales helps also. The NFL wants the teams in good markets who have more potential to get visibility and perform well.
4) Fanbase growth - How does the league grow a mature fanbase that is larger than any sport in the country? One way is by bringing new fans into games to watch two brothers play against each other who normally wouldn't care.
